what is the appropriate test method for testing the condition of this component when it is completely isolated

I need more information about what you're testing and what you're trying to evaluate.  IF it's a complicated testing/evaluation setup, a sketch or a diagram might be helpful.

I am trying to identify a defect in a returned board that was most likely hit with a power surge. if i test each component in a line from the line power the most likely breakage is where the power stops and i should be able to replace that and the P6KEdiode and the UF4005 diode and this should be the total sum of the damaged components? do you agree?

Many different components can fail during power surges.  Basic  troubleshooting rules apply here.  I would start by looking for visibly damaged components first.  Then work down through the parts most likely to get damaged next.  This would be parts that have voltage ratings likely to get exceeding during a power surge: diodes, ICs, caps, etc.
